| Background: | Streptavidin (SA) is a homotetramer protein secreted by Streptomyces avidinii during its growth. Like avidin, one mole of streptavidin can bind to four moles of biotin and has high affinity with biotin. Because SA does not contain sugar groups, it has a lower non-specific binding level than avidin in detection applications, and has been widely used in such biotechnology fields as enzyme-linked immunosorbent assay, immunohistochemistry, time resolved immunofluorescence (TRFIA), quantitative PCR, single strand DNA preparation, biological purification, and monoclonal antibody preparation. Compared with the natural streptavidin, the recombinant core streptavidin removes the sequence unrelated to the activity and only retains the core sequence related to the activity. Its stability and solubility are superior to the natural SA. In addition, cysteine is added at the C terminus, which can be covalently bound to the carrier. |
